Rotavirus is a major cause of acute diarrhea in both many kinds of young animals and children under 5 years old.Rotavirus NSP1, a 55 ku RNA binding protein, is the product of gene 5, which can subvert innate immune responses and be one of virulent determinant factors.According to the sequence in GenBank, specific primers targeting to NSP1 gene were designed and the gene was amplified by RT-PCR, following by being cloned into the pET-28a(+) vector.It showed that the full length of NSP1 gene was 1 473 bp, encoding 491 amino acids.The NSP1 shared the highest identity with WC3 strain.The recombinant protein was induced in E.coli Rosetta(DE3) by IPTG and was analyzed by SDS-PAGE and Western blotting.The results revealed that NSP1 recombinant protein existed in the form of inclusion body with the molecular weight of 55 ku.The purified recombinant protein could be recognized by His-tag antibody.This study laid the foundation for further research on the relationship between the intracytoplasmic location of NSP1 protein and its activity. 相似文献

【目的】利用木材动态压缩加载试验研究热磨机研磨解离破碎阶段木材原料受动态压缩载荷的动力学特性,并研究应变率、加载方向对木材原料动力学特性的影响,旨在深化木材原料研磨解离机制的研究,为热磨法纤维分离设备及磨片的齿形结构优化设计提供理论指导。【方法】利用 Hopkinson 杆试验装置对含水率为12.65%、密度为0.50 g·cm -3的桦木试件进行应变率约为400,800,1200 s -1,加载方向为径向、弦向和轴向的动态压缩试验,获得桦木在不同应变率及方向上动态压缩加载的解离特征、动态应力-应变曲线及相应的力学特性。【结果】对比分析各组试验后试件的破坏形态发现:1)当应变率约为400 s -1时,径向、弦向和轴向加载的试件主要发生塑性变形;2)当应变率约为800 s -1时,径向加载试件被解离成几大块,并且有一些“火柴棍”状的小试件从大试件上剥离;弦向加载试件沿加载方向上产生更大的塑性变形,并且在试件上、下两端处沿加载方向产生贯穿性裂纹,试件被解离成三大块;轴向加载试件受载面边缘处的纤维大量产生压溃现象,在加载面上产生贯穿性裂纹且有片状小试件从大试件上剥离;3)当应变率约为1200 s -1时,径向加载试件被解离成大量“火柴棍”状的小试件,并且小试件的尺寸明显小于应变率为800 s -1时;弦向加载试件沿加载方向上产生的塑性变形与应变率为800 s -1时相当,但是试件上端处被解离成许多片状的小试件,并且沿加载方向试件上产生了数条贯穿整个试件的大裂纹;轴向加载试件被解离成大量短粗状的小试件,并且小试件上带有明显的褶皱。对比分析各组试验试件的应力-应变曲线发现:1)动态压缩加载条件下桦木的应力-应变曲线可以由屈服点应变分为弹性阶段和屈服后弱线性强化阶段2部分;2)桦木沿径向加载应变率约为400,800,1200 s -1时,其屈服强度和韧性模量分别为4.56,10.49,14.22 MPa 和2.88,8.32,20.70 kJ·cm -3,应变率从400 s -1增加到1200 s -1时,屈服强度和韧性模量分别增加了2.11和6.19倍;3)桦木沿弦向加载应变率约为400,800,1200 s -1时,其屈服强度和韧性模量分别为5.87,7.90,9.65 MPa 和2.53,7.41,12.92 kJ·cm -3,应变率从400 s -1增加到1200 s -1时,其屈服强度和韧性模量分别增加了0.64和4.10倍;4)桦木沿轴向加载应变率约为400,800,1200 s -1时,其屈服强度分别为22.90,71.41,96.37 MPa 和18.79,67.74,114.32 kJ·cm -3,应变率从400 s -1增加到1200 s -1时,其屈服强度和韧性模量分别增加了3.21和5.08倍。【结论】随着应变率的增加,桦木的解离程度加大,径向加载最易解离,轴向加载最难解离;桦木的动态压缩屈服强度、动态压缩韧性模量具有很强的应变率敏感性,是一种应变率敏感材料。 相似文献

Duck meat consumption is low but steady in the United States (US), while rapidly expanding in other countries. The duck industry is moving from a traditional whole carcass product towards deboned breast fillets and other portions, which has prompted strain selection for breast weight and yield. The effect of bird strain was therefore evaluated for fillet weight, yield, and breast meat quality characteristics. Eighty butterflied paired fillets were obtained from each of 4 Pekin duck strains after carcasses were sized by weight at the processing plant (n = 640). All fillets were weighed and then half of the fillets were evaluated for color. One fillet from each carcass-pair was either frozen or kept fresh for cooking, reweighing, and subsequent color analysis (Commission Internationale de I'Eclairage (CIE) lightness L*, redness a*, and yellowness b*). Both fillets in each carcass-pair were evaluated using Allo–Kramer (AK) and Warner–Bratzler (WB) shear. Strain affected (P < 0.05) raw and cooked fillet weight. Strain C, with the largest BW and selected for both breast yield and growth rate, had higher raw and cooked breast weight. Cooked yield was not affected by strain and averaged 64.4%. Neither AK nor WB shear values were affected by strain, and averaged 4.8 kg/g and 4.2 kg, respectively. Strain had a slight but significant effect on color, affecting lightness (L*) values of all categories, and redness (a*) values of raw and cooked meat. Freezing and thawing produced large and significant thaw losses ranging from 10.4 to 13.6%; differing losses depended on the strain of duck. Duck strain affected breast fillet weights and lightness to a slight extent, but not cook yield or shear values. 相似文献

An 8‐week feeding experiment was conducted to determine the effect of dietary betaine levels on the growth performance and hepatic intermediary metabolism of genetically improved farmed tilapia (GIFT) strain of Nile tilapia Oreochromis niloticus (mean initial body weight: 78.3 ± 1.3 g, means ± SD). Six practical diets were formulated with the incorporation of betaine at the levels of 0 (control), 5, 10, 15, 20 and 25 g kg−1. Survival showed no significant differences among the treatments (P > 0.05). The highest and lowest weight gain (WG) and specific growth rate (SGR) were observed for fish fed the diets containing 5 and 0 g kg−1 (control) betaine, respectively. Feed intake showed similar trend with WG and SGR. In contrast, feed conversion ratio was the lowest when dietary betaine level was 5 g kg−1. In general, dietary betaine supplementation showed no significant effect on hepatic composition of tilapia. Condition factor and viscerosomatic index tended to increase with increasing dietary betaine levels from 0 to 5 g kg−1 and then decline when dietary betaine levels further increased from 5 to 25 g kg−1. In contrast, hepatosomatic index declined with increasing dietary betaine levels (P < 0.05). Dietary betaine levels significantly influenced several hepatic enzymatic activities, including succinate dehydrogenase, lactate dehydrogenase, malic dehydrogenase, lipoprotein lipase and hepatic lipase, suggesting that dietary betaine addition had significant effects on nutrient metabolism in the liver. Based on the second‐order polynomial regression analysis of WG, 12.5 g kg−1 of dietary betaine level seemed optimal for genetically improved farmed tilapia strain of O. niloticus. 相似文献

通过醋酸钠-抗生素筛选法从河北省土壤中筛选得到了苏云金芽孢杆菌新菌株CY1。镜检可观察到伴孢晶体呈小菱形。SDS-PAGE分析结果表明菌株CY1表达的晶体蛋白分子量为130 kDa。利用25对通用引物检测cry1,cry1I,cry7,cry2,cry3,cry4/cry10,cry5,cry6,cry7,cry8,cry9,cry11,cry13/14,cry16/17,cry18,cry19,cry21,cry22,cry24/25,cry26/28,cry27/29,cry30,cry32,cry34/35,cry40基因,仅从CY1菌株中检测出cry7基因,该cry7基因N末端的1 215 bp片段所编码的氨基酸序列与已发表的Cry7Ab1(Acc No.:U04367)序列同源性达99%,仅有2个氨基酸的差异。用3对cry7Ab特异引物检测,进一步证实该菌株携带有cry7Ab基因。 相似文献
